Description
Interleukin-8 (IL-8) belongs to the CXC chemokine family. This cytokine has a wide range of proinflammatory properties ranging from its main function as chemoattractant for neutrophils, basophils and a subpopulation of lymphocytes, to activation of neutrophils. This ELR motive (Glu-Leu-Arg motive in NH2 terminus) positive CXC chemokine is a potent activator of angiogenesis. In vivo, IL-8 is detected in conditions of inflammation. High levels of IL-8 are observed during septic episodes in seriously ill patients. Various observations strongly suggest a role for the CXC chemokines IL-8 and IP-10 in the regulation of angiogenic activity in cancer and in idiopathic pulmonary fibrosis. In vitro, multiple cell types can produce IL-8. The cytokine can be rapidly detected after activation of cells with a wide variety of activators; endotoxin is a potent inducer of IL-8 release by monocytes and endothelial cells. The human IL-8 ELISA kit shows cross reactivity with most old world monkeys.

Application
The Human IL-8 ELISA has been developed for the quantitative measurement of natural and recombinant human IL-8 in serum, plasma, and cell culture medium. In serum or plasma samples human IL-8 can be measured accurately if serum or plasma samples are diluted at least 2 times. Most reliable results are obtained if EDTA plasma is used.

Principle
- The human IL-8 ELISA is a ready-to-use solid-phase enzyme-linked immunosorbent assay based on the sandwich principle with a working time of 4½ hours.
- The efficient format of 2 plates with twelve disposable 8-well strips allows free choice of batch size for the assay.
- Samples and standards are captured by a solid bound specific antibody.
- Biotinylated tracer antibody will bind to captured human IL-8.
- Streptavidin-peroxidase conjugate will bind to the biotinylated tracer antibody.
- Streptavidin-peroxidase conjugate will react with the substrate, tetramethylbenzidine (TMB).
- The enzyme reaction is stopped by the addition of citric acid.
- The absorbance at 450 nm is measured with a spectrophotometer. A standard curve is obtained by plotting the absorbance (linear) versus the corresponding concentrations of the human IL-8 standards (log).
- The human IL-8 concentration of samples, which are run concurrently with the standards, can be determined from the standard curve.
Storage and stability
Product should be stored at 4°C. Under recommended storage conditions, product is stable for at least six months. After reconstitution the reagents are stable for 1 month if stored at 2-8°C.
