Collectins, Ficolins description
Collectins (collagen-like lectin) are proteins with collagen and lectin domains that play an important role in the mammalian first line host defense. Recent insights have clarified the structural basis of ligand recognition, the interactions of collectins with complement and the association with disease susceptibility. Well-known members of the collectin family are Mannose-Binding Lectin (MBL) and the lung surfactant proteins (SP-A and SP-D). Collectins bind to terminal non-reducing sugar residues, mannose, GlcNAc, fructose and glucose. MBL is able to activate the complement pathway independent of the classical and alternative complement activation pathways. The surfactant-associated proteins SP-A and SP-D are involved in the pulmonary innate immune response to microbial challenge. Ficolins are proteins, that consist of a collagen-like domain and a fibrinogen-like domain. Two types of ficolins, L-ficolin/P35 and H-ficolin (Hakata Ag), circulate in man. L-ficolin/P35 is associated with Mannose-binding lectin (MBL)-associated serine proteases (MASP) forming a complex that can activate the lectin pathway. HB offers antibodies and assays to human MBL, SP-D, unique assays for mouse MBL-A and MBL-C, L-ficolin, H-ficolin, MASP2, and MBL/MASP2.
