Short description
The classical pathway of complement and the Mannose binding lectin activation pathway converge at C4. C1s, MASP-1 and MASP-2 cleave C4 resulting in the formation of C4a and C4b. The latter can be cleaved to result in C4c and C4d in which step all functional sites are lost. The monoclonal antibody 16D2 reacts with intact C4, C4b and C4d. C4 is an acute phase protein that is produced by hepatocytes, monocytes and intestinal epithelial cells.

Formulation
0.25 ml of approximately 1 mg/ml purified natural human C4 isolated from healthy blood donors, in Phosphate buffered saline, pH 7.2.The blood donors have been tested and found negative for various viruses (see table: “Human blood test results”). The exact protein concentration and functional activity is indicated on the label.

Application notes
The biological activity of C4 is defined as the amount of C4 required to yield 50% lysis of sensitized erythrocytes (EA).
Use
For functional studies, in vitro dilutions have to be optimized in user’s experimental setting.
Storage and stability
Product should be stored at –70°C. Repeated freeze and thaw cycles will cause loss of activity. The exact expiry date is indicated on the label. Use C4 protein within 24 hours after thawing and keep on ice. Remainder amounts should be aliquoted and immediately re-freezed for future use. Aliquots should never be thawed more than once. Under recommended storage conditions, product is stable for at least one year.
