Description
LPS binding protein (LBP) is an approximately 60 kDa acute phase protein that is produced by hepatocytes. This protein strongly binds to LPS and has been shown to play an important role in the handling of LPS by the host. A number of functions of LBP have been reported. First, LBP transfers LPS to the LPS receptor CD14 on mononuclear phagocytes, leading to an 100-1,000-fold increased sensitivity of the cells to LPS. Furthermore, LBP can enhance the response of CD14 negative cells by acceleration of LPS binding to soluble CD14, a complex that stimulates these cells. Next, LBP transfers LPS into High Density Lipoprotein (HDL) which effectively neutralizes its biological potency. LBP was demonstrated to protect mice from septic shock caused by LPS or gram negative bacteria.
Formulation
1 ml of approximately 10 µg/ml purified natural LBP in PBS. The exact protein concentration is indicated on the label.
Species
Human LBP is obtained by human plasma purification. The plasma used has been tested and found negative for various viruses (see table: "Human plasma test results"). However handle human LBP according to guidelines for preventing transmission of blood-borne infections.
| Human plasma test results |
| HBsAg | negative |
| HCV | negative |
| Anti-HIV 1 and HIV 2 | negative |
| VDRL | negative |
Use
For dilutions use protein stabilized phosphate buffered saline, pH 7. (Never use fresh serum/plasma.) Be aware: endotoxin will bind LBP.
Storage and stability
The stock solution should be stored in aliquots at -70°C in polypropylene tubes. LBP stock solution should never be thawed more than once. Repeated freeze and thaw cycles will cause loss of activity. Under recommended storage conditions, product is stable for one year.
