Introduction sCD14 (Cat.# HK320)

The protein encoded by this gene is a component of the innate immune system. CD14 exists in two forms. Either it is anchored into the membrane by a glycosylphosphatidylinositol tail (mCD14) or it appears in a soluble form (sCD14). Soluble CD14 either appears after shedding of mCD14 (48 KDa) or is directly secreted from intracellular vesicles (56 KDa). CD14 takes its name from its inclusion in the cluster of differentiation group of cell surface marker proteins. CD14 was the first described pattern recognition receptor. CD14 acts as a co-receptor (along with the Toll-like receptor TLR 4 and MD-2) for the detection of bacterial lipopolysaccharide (LPS). CD14 can bind LPS only in the presence of lipopolysaccharide-binding protein (LBP). Although LPS is considered its main ligand, CD14 also recognizes other pathogen-associated molecular patterns. CD14 is expressed mainly by macrophages and (at 10 times lesser extent) by neutrophil granulocytes. It is also expressed by dendritic cells. A soluble form sCD14 is secreted by the liver and monocytes and is sufficient in low concentrations to confer LPS-responsiveness to cells that otherwise do not express CD14. sCD14 is also present in human milk, where it is believed to regulate microbial growth in the infant gut. CD14 has been shown to interact with Lipopolysaccharide-binding protein.

LBP is a soluble acute-phase protein that binds to bacterial lipopolysaccharide (or LPS) to elicit immune responses by presenting the LPS to important cell surface pattern recognition receptors called CD14 and TLR4. The protein encoded by this gene is involved in the acute-phase immunologic response to gram-negative bacterial infections. Gram-negative bacteria contain a glycolipid, lipopolysaccharide (LPS), on their outer cell wall. Together with bactericidal permeability-increasing protein (BPI), the encoded protein binds LPS and interacts with the CD14 receptor, probably playing a role in regulating LPS-dependent monocyte responses. Studies in mice suggest that the encoded protein is necessary for the rapid acute-phase response to LPS but not for the clearance of LPS from circulation. This protein is part of a family of structurally and functionally related proteins, including BPI, plasma cholesteryl ester transfer protein (CETP), and phospholipid transfer protein (PLTP). Finally, this gene is found on chromosome 20, immediately downstream of the BPI gene. Lipopolysaccharide-binding protein has been shown to interact with CD14, TLR2, TLR4 and the co-receptor MD-2. The host response to bacterial compounds is regulated and modulated by certain serum proteins and cellular receptor molecules. LBP and soluble CD14, both able to bind LPS, are present in serum in high quantities. LBP is an acute-phase protein synthesized in the liver, the concentration of which rises dramatically during systemic infection and the acute-phase response. It monomerizes LPS vesicles and transports LPS to the CD14 part of the cellular LPS receptor enabling inflammatory responses, such as TNF-α synthesis. LPS effects can be blocked by a range of inhibitors including polymyxin B, a polypeptide known to bind lipid A, the active moiety of LPS.
