Description
The monoclonal antibody ED31 recognizes the macrophage receptor with collagenous structure (MARCO). The scavenger receptors (SRs) expressed by macrophages are thought to play an important role in the immune response against bacteria by mediating binding and phagocytosis. An important family of receptors involved in these processes are the class A macrophage scavenger receptors (SR-A). There are three classes of SRs, A, B and C. SR-A, type I and type II, are homotrimeric glycoproteins composed of three 77 kD monomers. MARCO is a member of this class of scavenger receptors. The molecular structure of MARCO resembles that of SR-AI, containing a triple-helical collagenous domain and a scavenger receptor cysteine-rich (SRCR) domain at the C terminus. MARCO is expressed by distinct populations of macrophages in the spleen and lymph nodes. It is rapidly induced on macrophages in other tissues (e.g. Liver Kupfer cells) during infection or LPS treatment. MARCO seems to play an important role in the lungs response to inhaled particles and airborne pathogens. The monoclonal antibody ED31 binds to the C-terminal cysteine rich domain of MARCO, and has been shown to block ligand binding.
Formulation
1 ml (100 µg/ml) 0.2 µm filtered antibody solution in PBS, containing and 0.1% bovine serum albumin.
Species
Rat IgG1
Application
The monoclonal antibody ED31 can be used for immunohistology on frozen sections. Furthermore the monoclonal antibody ED31 is useful for flow cytometry, immuno precipitation and neutralization of biological activity.
Use
For immunohistology and flow cytometry dilutions to be used depend on detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:50. For neutralization of biological activity in vitro dilutions have to be made according to the amounts of MARCO to be inactivated.
Storage and stability
Product should be stored at 4°C. Under recommended storage conditions, product is stable for one year.
