Hycult Dx Complement factor H ELISA

The Hycult Dx Complement factor H ELISA (Cat.# DH003) is a new ELISA in the diagnostic product range of Hycult Biotech, only available in EU. Complement factor H (CFH) is one of the most abundant plasma proteins with a concentration of 300-800 mg/l. CFH a protein in the alternative pathway of the complement system and is essential to maintain homeostasis and restrict the action of complement to activating surfaces.

Related to the regulatory function of CFH, deficiency or dysfunction can lead to a higher susceptibility to diseases as Haemolytic Uremic Syndrome (HUS) and infectious diseases.

Specifications of the assay:

-       Concentration range of 3.9 – 250 ng/ml
-       Detection limit of 3.9 ng/ml
-       Useful for serum, plasma and urine samples
-       Useful for 40 samples in duplicate / 80 samples single
-       Positive and negative control included
-       Working time of 2.5 hrs
-       No cross reactivity with mouse, rat, bovine and horse

Linearity

The linearity of the assay was determined by serially diluting a sample containing 250 ng/ml human factor H. The diluted samples were measured in the assay. The line obtained a slope of 0.9982 and a correlation coefficient of 1.

Precision and reproducibility

Four human plasma samples with known human factor H concentration were tested in 4 replicates within 4 runs (intra-assays),  and in 4 replicates in 4 separate runs (inter-assay).

The intra-assay precision and reproducibility was tested with samples of three different, known concentrations in 6 replicates, performed by 3 different operators.

The inter-assay precision and reproducibility was tested with samples of three different, known concentrations, assayed in duplicate in 6 separate runs. These were performed by 3 different operators: single run per operator on 2 different days.

 Recovery

Normal human blood samples (plasma) containing baseline levels of human factor H, were spiked with human factor H  in concentrations of 50 and 200 ng/ml. Samples with and without human factor H were incubated for 1 hour at room temperature. Samples were measured using the ELISA. Values for human factor H ranged between 90.6% and 101.5% (mean 93.9%).
