anti-FHR-2; clone 2.12 is a mouse monoclonal antibody recognizing human full length Complement Factor H-related protein 2 (FHR-2), a glycoprotein belonging to the family of complement regulatory proteins. FHR-2 has a molecular weight of approximately 29 kDa and is synthesized and secreted primarily by the liver but also by various immune cells, including monocytes, macrophages, and dendritic cells. Structurally, FHR-2 shares homology with Complement Factor H (CFH), which is a key regulatory protein within the complement system. Also FHR-2 is involved in the regulation of the complement cascade but its biological role is poorly understood somewhat controversial. Studies showed that FHR-2 inhibits the activity of the complement alternative pathway (AP) C3 convertase, thereby preventing MAC formation. On the other hand, it is described that a hybrid mutant protein consisting of FHR-5 domains and full length FHR-2 causes complement overactivation, which suggests that FHR-2 rather promotes AP activation. Dysregulation of FHR-2 is associated with diseases such as atypical hemolytic uremic syndrome (aHUS) and age-related macular degeneration (AMD). In these diseases, abnormal complement activation and deposition on host tissues can contribute to pathological processes. FHR-2’s involvement in the alternative pathway of the complement system positions it as a potential target for therapeutic interventions aimed at modulating complement-related diseases
